Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
CD16 (Fc gamma III Receptor) and CD32 (Fc gamma II Receptor) are low-affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
For use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤ 1.0 µg/test.
A test = antibody amount for 100 µL final volume (10⁵–10⁸ cells/test).
Antibody titration recommended for optimal performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures, providing optimized fluorescence with reduced spillover and spread.

Target Information

CD16:
A 50–65 kDa surface molecule existing in two forms:

• Transmembrane (NK cells, some T cells)
• Phosphatidylinositol-linked (granulocytes)
  Functions as a low-affinity receptor for IgG (FcR III) and mediates antibody-dependent cell-mediated cytotoxicity (ADCC).

CD32:
A 40 kDa glycoprotein, low-affinity receptor for IgG (Fc gamma RII).
Mediates endocytosis, secretion activation, cytotoxicity, and immunomodulation.
Expressed by B cells, monocytes, granulocytes, and platelets.
Involved in immune complex phagocytosis and regulation of antibody production.
Variants may increase susceptibility to systemic lupus erythematosus (SLE).
