Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II molecules and is crucial for T-cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested with mouse thymocytes and splenocytes at ≤0.5 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

NovaFluor Dyes Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

Buffer Information:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Note:

Target Information

CD4 is a co-receptor for MHC class II recognition and HIV binding. Expressed mainly in T-helper lymphocytes, monocytes, macrophages, and dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 aids T-cell activation via association with the T-cell receptor complex and Lck kinase. Also implicated in neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
