Product Specific Information
Specificity:
AHT107 binds to a different epitope than IL-2. The determinant recognized by AHT107 is involved in formation of the high-affinity binding site of the p55 + p75 heterodimeric complex of the IL-2R, without disturbing the interaction of the p55 molecule and IL-2. The high-affinity class of the IL-2R mediates the growth signal at physiological concentrations of IL-2.
Preparation:
BALB/c mice were immunized with human T lymphoblasts, and spleen cells from the immunized animals were fused with X63-Ag8.653 mouse myeloma cells. The supernatants of the resultant hybridomas were screened by a proliferation inhibition assay (inhibition of the human T lymphoblast response to IL-2). Two clones (AHT54 and AHT107) that produced antibodies inhibiting IL-2 dependent proliferation of human T lymphoblasts were selected, cloned, and recloned.

CD25 (IL2 receptor alpha chain / IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation.
The IL2RA receptor forms a heterotrimeric complex with IL2RB and IL2RG, constituting the high-affinity IL2 receptor.
- Homodimeric IL2RA → low-affinity receptor
- Homodimeric IL2RB → medium-affinity receptor
IL2 is primarily produced by mature T cells and functions as a growth, differentiation, and apoptosis regulatory factor. It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation.
Disruption of the IL2RA gene in mice causes ulcerative colitis-like disease, indicating its role in immune response regulation.
Mutations in this gene are associated with interleukin 2 receptor alpha deficiency.
