Specificity:
AHT107 binds to a different epitope than IL-2. The determinant recognized by AHT107 is involved in forming the high-affinity binding site of the p55 + p75 heterodimeric complex of the IL-2R without disturbing the interaction between p55 and IL-2. The high-affinity IL-2R class mediates growth signaling at physiological IL-2 concentrations.
Preparation:
BALB/c mice were immunized with human T lymphoblasts, and spleen cells were fused with X63-Ag8.653 mouse myeloma cells. Supernatants of hybridomas were screened by inhibition assays for IL-2 dependent proliferation. Clones AHT54 and AHT107 were selected, cloned, and recloned for further use.
Target Information
CD25 (IL2 receptor alpha chain/IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2RA forms part of a heterotrimeric receptor complex with IL2RB and IL2RG, constituting the high-affinity IL2 receptor.
- Homodimeric IL2RA → low-affinity receptor
- Homodimeric IL2RB → medium-affinity receptor
IL2 expression is monoallelic in mature thymocytes, regulating precise gene expression. IL2, produced mainly by mature T cells, acts as a growth, differentiation, and cell death regulator. It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation. Mouse models with disrupted IL2RA genes develop ulcerative colitis-like symptoms, indicating its role in immune responses. Mutations in IL2RA are associated with interleukin 2 receptor alpha deficiency.
