Target Information

TRPM4 is a calcium-activated, non-selective cation channel that mediates membrane depolarization. Although activated by increased intracellular Ca²⁺, it is impermeable to Ca²⁺ itself. It transports monovalent cations (Na⁺, K⁺, Cs⁺, Li⁺), contributing to depolarization of the membrane.
It plays a central role in cardiomyocytes, neurons, endocrine pancreas cells, kidney epithelial cells, cochlea hair cells, and more. TRPM4 participates in T-cell activation by modulating Ca²⁺ oscillations required for NFAT-dependent IL2 production, and is involved in myogenic constriction of cerebral arteries.
Additionally, it controls insulin secretion in pancreatic beta-cells and may affect pacemaking or irregular electrical activity under Ca²⁺ overload. TRPM4 influences T-helper cell motility and cytokine production via calcium signaling and NFATC1 localization, and enhances cell proliferation through β-catenin signaling upregulation.
