Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is tonsil tissue.
Target Information
CD22 (BL-CAM) is a type 1 integral membrane glycoprotein with a molecular weight of 130–140 kDa.
It is expressed in both the cytoplasm and cell membrane of B-lymphocytes and appears early in B-cell differentiation, similar to CD19.
CD22 membrane expression is limited to mature B cells (CD22+) and plasma cells (CD22-), making it useful for phenotyping mature leukemias.
CD22 is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
It binds preferentially to α2,6-linked sialic acid and plays roles in both positive and inhibitory regulation of B-cell receptor signaling through tyrosine phosphorylation and phosphatase recruitment.
