Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Applications Reported:
Use in flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry on normal human peripheral blood cells. Recommended usage: 5 µL (0.03 µg)/test. A test is defined as staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Fluorophore Information:
Alexa Fluor® 532 is excited with the Green laser (532 nm) and emits at 561 nm. Not compatible with the Yellow-Green laser (561 nm). Recommended filter: 560/14 band pass.

Target Information

The CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed in T helper cells, with additional expression in monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 aids early T-cell activation via interaction with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
