Product Specific Information

Description:
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is critical for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤0.5 µg per test (100 µL final volume). Optimal titration is advised for each assay. Typical cell count range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Alexa Fluor 532 is excited with the Green laser (532 nm) and emits at 561 nm. It is not compatible with the Yellow-Green laser (561 nm). Recommended filter: 560/14 band pass. Confirm instrument compatibility before use.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV, expressed mainly on T helper cells, and also on monocytes, macrophages, and dendritic cells. It is found in thymus, lymph nodes, tonsils, spleen, and certain regions of the brain and gut. CD4 participates in T-cell activation via association with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
