Product Specific Information

The A2MR-a2 monoclonal antibody reacts with human CD91 (also known as LRP-1 or alpha-2 macroglobulin receptor). CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains.

• The alpha chain binds multiple ligands; the beta chain contains tyrosine phosphorylation motifs for adaptor and signaling protein interactions.
• Expressed in monocytic cells, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, and syncytiotrophoblasts.
• Involved in antigen cross-presentation via MHC class I, cell migration, and signaling activation.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended use: 5 µL (0.4 µg) per 100 µL test volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence with minimal spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

The encoded protein is an endocytic receptor involved in intracellular signaling, lipid homeostasis, and apoptotic cell clearance. It is essential for A2M-mediated clearance of amyloid precursor protein and beta-amyloid, key components of Alzheimer’s plaques. Expression decreases with age and is reduced in Alzheimer’s brain tissue.
