Product Specific Information
The A2MR-a2 monoclonal antibody reacts with human CD91 (LRP-1, alpha-2 macroglobulin receptor). CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains. The alpha chain binds multiple ligands, while the beta chain includes tyrosine phosphorylation motifs interacting with adaptor and signaling proteins. Ligand binding mediates endocytosis and lysosomal trafficking.
CD91 is expressed by monocytes, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, and syncytiotrophoblasts. It participates in antigen cross-presentation via MHC class I and plays roles in cell migration and signaling activation.

Applications
- Reported for use in flow cytometric analysis
- Pre-titrated and tested on normal human peripheral blood cells
- Recommended use: 5 µL (0.05 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)
Important Notes
- NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
- Add 5 µL per sample (regardless of number of NovaFluors).
- For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescence with minimal spillover and spread.

Target Information
The encoded protein functions as an endocytic receptor involved in intracellular signaling, lipid homeostasis, and apoptotic cell clearance. It is required for A2M-mediated clearance of amyloid precursor protein and beta-amyloid, key components of amyloid plaques in Alzheimer’s disease. Expression decreases with age and is reduced in Alzheimer patient brain tissue.
