Product Specific Information

The RPA-2.10 monoclonal antibody reacts with human CD2, a 50 kDa cell surface receptor expressed by most thymocytes, all mature T cells, and a subset of NK cells. CD2 acts as a ligand for CD58 and plays a role in T cell adhesion and activation. The RPA-2.10 clone blocks mixed lymphocyte reactions and cross-reacts with non-human primates and pigs.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral blood cells at ≤ 5 µL (0.1 µg) per test.
A test is defined as the antibody amount required to stain a cell sample in 100 µL final volume (10⁵–10⁸ cells/test). Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample, regardless of the number of NovaFluors in the panel.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures engineered for optimized fluorescent performance and reduced spillover.

Target Information

CD2 (LFA-2) is a monomeric surface antigen (45–58 kDa) of the human T-lymphocyte lineage.

• Expressed on >95% of thymocytes and peripheral T cells
• Also found on some NK cells, but not on B lymphocytes
• Mediates adhesion via interaction with CD58, stabilizing T cell–APC contact
• CD2 expression increases upon T cell activation, enhancing adhesion and signaling
• CD2 deficiency reduces intestinal inflammation and aids infection control
• Dysregulation associated with immune deficiencies and certain carcinomas
