Target Information
STAT4는 다른 STAT 단백질의 보존된 영역을 인코딩하는 서열에 상보적인 퇴화 프라이머를 사용하여 처음 확인되었습니다. STAT4 단백질은 STAT1과 52%, STAT3과 47%의 유사성을 보입니다. 기능적으로 STAT4는 Jak1 또는 Jak2에 의해 티로신 인산화될 수 있으며, 동종이합체 및 이종이합체를 형성합니다. 인산화된 STAT4는 IFN-γ 활성화 부위(GAS)에 결합할 수 있으며, 세린 인산화도 최대 전사 활성에 필요합니다. STAT4 발현은 흉선, 비장, 고환에 제한됩니다. STAT4는 인터루킨-12(IL-12)에 의해 활성화되며, IL-12는 IFN-γ 유도 및 Th1 반응 발달에 필수적입니다. STAT4 결핍 생쥐는 Th1 세포 발달이 저하되고 Th2 세포 발달이 증가하는 특징을 보입니다. 또한 바이러스 감염 시 IFN-α/β에 의한 STAT4 활성화는 IFN-γ 생성에 필요합니다.
