Target Information
Peroxisome proliferators are non-genotoxic carcinogens that exert their cellular effects through interaction with nuclear hormone receptors known as peroxisome proliferator-activated receptors (PPARs). These ligand-dependent intracellular proteins stimulate transcription of specific genes by binding to DNA sequences upon activation by appropriate ligands.
PPARs are activated by compounds such as clofibric acid, nafenopin, WY-14,643, and certain fatty acids. They can induce transcription of acyl coenzyme A oxidase and cytochrome P450 (CYP450) A6 through interaction with specific response elements. PPARs, similar to other nuclear hormone receptors, heterodimerize with retinoid X receptor (RXR) alpha.
