Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.1 µg) per test (100 µL final volume).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails prior to cell mixing.

NovaFluor conjugates utilize Phiton technology based on nucleic acid dye structures, engineered for optimized spectral properties and minimal spillover.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and serves as a co-receptor for HIV. Expressed mainly on T helper cells, and also on monocytes, macrophages, and dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 aids T-cell activation via association with TCR and Lck kinase. It may mediate neuronal damage in infectious and immune CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
