Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV. SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Suitable for flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
• Use 5 µL (0.1 µg) per test (100 µL total volume).
• Cell count: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL per sample, regardless of the number of NovaFluors in the panel.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology based on novel nucleic acid dye structures, engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD4 antigen is involved in MHC class II molecule recognition and acts as a co-receptor for HIV. It is primarily expressed on T helper cells and also on monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a role in early T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
