Target Information

STAT3 (Signal Transducer and Activator of Transcription 3) is a member of the STAT protein family. It is phosphorylated by receptor-associated kinases and translocates to the nucleus to act as a transcription factor in response to cytokines and growth factors. Coactivators such as CREB-binding protein are necessary for transcriptional activation by STAT3.

STAT3 can be activated by Interferon-alpha, Interferon-gamma, EGF, PDGF, and IL-6. Phosphorylation on Tyr705 by JAK1 and JAK2 is critical for dimer formation, nuclear translocation, and DNA binding. The human STAT3 gene is located on chromosome 17q. It is activated by IFN-alpha but not IFN-beta and interacts with transcription factors such as c-Jun and CREB.

STAT3 regulates genes involved in cell growth and apoptosis, playing a key role in various cellular processes. Rac1 GTPase binds and regulates STAT3 activity, while PIAS3 acts as a specific inhibitor. Three alternatively spliced transcript variants encoding distinct isoforms have been identified. Deletion of STAT3 in knockout mice results in embryonic lethality.
