Product Specific Information

The FNLAP monoclonal antibody reacts with human latency associated peptide (LAP, pro-TGF beta 1, LAP/TGF beta 1).
TGF beta is synthesized as a precursor containing LAP at the N-terminus and mature TGF beta at the C-terminus.
Processing between amino acids 278 and 279 forms LAP and TGF beta dimers, which associate to create the latent TGF beta complex.
LAP is secreted and found in the extracellular matrix and can also be expressed on platelets and activated regulatory T cells due to binding with GARP (LRRC32).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry using stimulated human peripheral blood cells and Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) following BestProtocols® Protocol B.

Use at 5 µL (0.5 µg)/test in 100 µL final volume.
Cell number may range from 10⁵ to 10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

TGF beta-1 is a member of the transforming growth factor beta superfamily, widely found in tissues.
It is stored in the extracellular matrix as a latent complex with its prodomain.
Activation requires binding of aV integrin to an RGD sequence and mechanical force applied to the prodomain, which is anchored by latent TGF-beta binding proteins.
Latent forms are complexes of TGF-beta, the N-terminal portion designated TGF-LAP, and a specific binding protein LTBP.
