Product Specific Information

Description:
The V3GYWCH monoclonal antibody reacts with human and mouse interferon regulatory factor 8 (IRF8; ICSBP). IRF8 is a 50 kDa transcription factor that plays a critical role in dendritic cell (DC) development, particularly in plasmacytoid DC (pDC) and CD8a+ DC differentiation. IRF8-/- mice lack both pDC and CD8a+ DC populations, while CD8a- DCs remain but show impaired maturation and function. Retroviral IRF8 transduction restores pDC development from DC progenitors in IRF8-/- mice.

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.
Refer to BestProtocols®: Protocol B for intracellular (nuclear) proteins.
Recommended use ≤ 0.25 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

IRF8 (Interferon Regulatory Factor 8)
IRF8 is a transcriptional activator/repressor that binds to upstream regulatory regions of type I IFN and IFN-inducible MHC class I genes. It negatively regulates immune system cells and is involved in CD8+ dendritic cell differentiation via complex formation with BATF-JUNB heterodimer. This complex recognizes the AICE sequence, promoting cooperative binding and gene activation. Mutations in IRF8 are associated with immunodeficiency types 32A and 32B.
