This HOST5DS monoclonal antibody recognizes human GPNMB, also known as Osteoactivin, DC-HIL, and HGFIN.
GPNMB is a type-1 transmembrane glycoprotein involved in bone development, tumor progression, neurokinin signaling, and immune regulation. It is expressed on macrophages and dendritic cells and interacts with Syndecan-4 (SD-4) on T cells to inhibit their activation and proliferation.

Surface expression of GPNMB on myeloid cells may indicate tolerogenic properties. Various cancers express GPNMB, aiding in immune evasion. Expression is upregulated by IL-10 and immunosuppressive drugs such as dexamethasone. THP-1 and U937 cell lines constitutively express GPNMB, which may also be secreted.

The HOST5DS antibody is effective on paraformaldehyde-fixed and permeabilized cells and suitable for intracellular staining. Note that enzymatic tissue dissociation may reduce surface GPNMB levels and staining brightness.

Target Information

GPNMB is a type I transmembrane glycoprotein homologous to pMEL17, a melanocyte-specific protein. It is expressed in lowly metastatic melanoma cell lines but not in highly metastatic ones, suggesting a role in growth delay and reduced metastatic potential. Two transcript variants encoding different isoforms have been identified.
