Product Specific Information

Description:
The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except erythrocytes and platelets. It has tyrosine phosphatase activity, critical for lymphocyte activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended 5 µL (0.25 µg) per 100 µL cell sample.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is used with a 355 nm ultraviolet laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Compatible with short-term methanol-based fixation (≤1 hour).

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues and regulates Src-family kinases, T/B-cell receptor signaling, cytokine receptor signaling, and integrin-mediated pathways. CD45 isoforms arise from alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions. It plays essential roles in immune cell activation, signaling, and survival.
