Target Information
KLOTHO is a systemic anti-aging hormone within the glycosidase 1 superfamily. It encodes a type I membrane protein abundant in the kidney and brain.
In mice, KLOTHO deficiency leads to phenotypes resembling human aging, such as arteriosclerosis, osteoporosis, and skin atrophy, along with growth retardation, shortened lifespan, and infertility.
Overexpression of KLOTHO extends lifespan by inhibiting insulin/IGF1 signaling.
It also regulates calcium/phosphorus homeostasis by inhibiting active vitamin D synthesis and is identified as a potential tumor suppressor.
