Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing mouse strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

Applications Reported:
The A20 antibody is suitable for flow cytometric analysis, immunoprecipitation, and immunohistochemical staining of frozen tissue sections. (Biotinylated A20 recommended for IHC; Product #13-0453.)

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤0.5 µg per test (defined as staining a 100 µL cell sample). Cell number may vary from 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It plays a key role in lymphocyte development and antigen signaling by regulating Src-family kinases.

CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains while sharing identical transmembrane and cytoplasmic regions. Isoform expression varies by cell type and physiological state.

CD45 also modulates integrin-mediated signal transduction, promotes cell survival, and participates in DNA fragmentation during apoptosis. CD45RA is an isoform with restricted expression among lymphoid subtypes.
