Product Specific Information

Description:
The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes, and B cells. CD45RA+ T cells are mainly naive/virgin, allowing use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Applications Reported:
Reported for use in flow cytometric analysis and immunohistochemical staining.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD45RA is an isoform of the CD45 complex, expressed variably among lymphoid cell subtypes. CD45 (leukocyte common antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins. It plays a critical role in lymphocyte development, antigen signaling, and regulation of Src-family kinases.

CD45 isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. They vary in membrane localization and expression depending on cell type and physiological state. CD45 also promotes cell survival through integrin-mediated signaling and is involved in DNA fragmentation during apoptosis. It is expressed on hematopoietic lineage cells except mature red cells and is absent in non-hematopoietic tissues.
