• For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell–cell junctions in endothelial cells and in other normal and malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may act as a signal transduction molecule through recruitment of Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a role in cell adhesion and cohesion of the endothelial monolayer in vascular tissue. CD146 expression may facilitate melanoma cell interaction with vascular elements, enhancing tumor spread. It may also function as an adhesion molecule in neural crest cells during embryonic development and as a receptor triggering tyrosine phosphorylation of FYN and PTK2, with a transient increase in intracellular calcium concentration.
