• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily.
It is expressed at cell-cell junctions in all endothelial cells and in other normal and malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may act in signal transduction through Fyn kinase activation and tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a role in endothelial cohesion and melanoma cell interaction with vascular elements, potentially enhancing tumor spread.
Additionally, CD146 may function as an adhesion molecule in neural crest cells during embryonic development and as a surface receptor triggering phosphorylation of FYN and PTK2 with transient intracellular calcium increase.
