Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal and malignant cell types. Beyond its role as a calcium-independent adhesion molecule, CD146 may function in signal transduction through recruitment of Fyn kinase and subsequent tyrosine phosphorylation of proteins involved in actin cytoskeleton assembly.
CD146 contributes to endothelial monolayer cohesion and may facilitate melanoma cell interaction with vascular components, promoting tumor spread. It also plays roles in neural crest cell adhesion during embryonic development and acts as a surface receptor influencing intracellular calcium signaling.
