Reconstitution:

• Briefly centrifuge vial before opening.
• Reconstitute leptin in sterile, distilled water to a concentration of 1.0–5.0 mg/mL.
• Do not vortex.
• Store solution at 2°C–8°C for up to 1 week.
• For extended storage, dilute further in buffer containing 0.1% BSA and store aliquots at -20°C to -80°C.

Storage:

• Lyophilized protein stable at room temperature for 1 month.
• For long-term storage, keep at -20°C.
• Working aliquots with carrier protein stable for ≥3 months at -20°C to -80°C.
• Avoid repeated freeze/thaw cycles.

Target Information

Leptin is a 167 amino acid long protein hormone with key roles in body weight regulation, metabolism, and reproductive function. It is encoded by the obese (ob) gene and primarily expressed in adipocytes, with smaller amounts secreted by stomach epithelium and placenta cells.

Leptin receptors are highly expressed in hypothalamic regions controlling hunger, body temperature, and energy expenditure, as well as in T lymphocytes and vascular endothelial cells.

• Food intake and energy expenditure
• Adiposity and thermogenesis
• Hematopoiesis, angiogenesis, wound healing, inflammation, immune responses
• Pubertal development and fetal growth

Defects in leptin production can lead to severe hereditary obesity in rodents and humans.
