Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone, but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). Binding of IL-2 to both receptor classes is inhibited by the PC61.5 antibody. CD25 is involved in lymphocyte differentiation and activation/proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Optimal dilution should be determined empirically (10⁵–10⁸ cells/test).
Fluorescence: Excitation 633–647 nm; Emission 660 nm; Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD25 (IL2 receptor alpha chain / IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2RA gene encodes the α-chain of the heterotrimeric IL-2 receptor complex (with IL2RB and IL2RG).

• Homodimeric IL2RA: low-affinity receptor
• Homodimeric IL2RB: medium-affinity receptor
• Heterotrimeric IL2RA/IL2RB/IL2RG: high-affinity receptor

IL2 is primarily produced by mature T cells and functions as a growth and differentiation factor, as well as a regulator of cell death. It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte-macrophage colony formation. Mutations in IL2RA are linked to interleukin-2 receptor alpha deficiency and immune dysregulation.
