Target Information

BK Virus VP1 forms an icosahedral capsid with T=7 symmetry and a 50 nm diameter. The capsid is composed of 72 pentamers linked by disulfide bonds and associated with VP2 or VP3 proteins. It interacts with gangliosides GT1b and GD1b containing terminal α2-8-linked sialic acids on the cell surface, enabling virion attachment to target cells. This attachment induces internalization mainly through caveolin-mediated endocytosis, followed by trafficking to the endoplasmic reticulum (ER). Inside the ER, the protein folding machinery isomerizes VP1 interpentamer disulfide bonds, triggering initial uncoating. The virion then likely uses ER-associated degradation machinery to translocate into the cytosol before reaching the nucleus. Nuclear entry involves selective exposure and importin recognition of VP2/VP3 nuclear localization signals. During the late phase of infection, newly synthesized VP1 encapsulates replicated genomic DNA in the nucleus and assists in rearranging nucleosomes around the viral DNA.
