Product Specific Information

Description:
The HIT2 monoclonal antibody reacts with human CD38, an approximately 45 kDa type II transmembrane protein expressed on thymocytes, peripheral B cells (including plasma cells), activated T cells, and monocytes. CD38 serves as a counter-receptor for CD31 and exhibits both cyclase and hydrolase enzymatic activity.

Applications Reported:
This HIT2 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested in flow cytometric analysis of normal human peripheral blood cells. Recommended usage is ≤0.5 µg per test (defined as the amount of antibody that stains a cell sample in 100 µL final volume). Optimal cell number ranges from 10⁵ to 10⁸ cells/test and should be empirically determined. Antibody titration is recommended for best performance.

Target Information

CD38 (NAD⁺ glycohydrolase):
CD38 is a type II transmembrane glycoprotein involved in activation, proliferation, and differentiation of lymphocytes, as well as apoptosis of progenitor cells. It functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, and NAD⁺ glycohydrolase) and as a cell surface receptor.

Antibodies to CD38 are useful for:

• Subtyping lymphomas and leukemias
• Detecting plasma cells (e.g., myeloma identification)
• Serving as markers for activated B and T cells

CD38 participates in cell adhesion, signal transduction, and calcium signaling, and is highly expressed in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Diseases associated with CD38 dysfunction include chronic lymphocytic leukemia and Richter’s Syndrome.
