Target Information
Connexin 43 (Cx43) is a member of the gap junction protein family. Connexins assemble as hexamers and are transported to the plasma membrane to form hemichannels that connect with adjacent cells to create cell-to-cell communication channels. These clusters form gap junctions, which are essential for development and regulation of cell growth.
Phosphorylation of Cx43, particularly at Ser368 by protein kinase C (PKC), regulates gap junction assembly and function by reducing intercellular communication. Src kinase can also phosphorylate Cx43, altering communication between cells. Connexin 43 is the predominant gap junction protein in the heart, contributing to synchronized cardiac contraction and embryonic development. Mutations in related genes can lead to disorders such as Charcot-Marie-Tooth disease and oculodentodigital dysplasia.
