Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed on thymocytes and mature T cells. Binding of 145-2C11 to TCR induces intracellular signaling that can lead to activation, proliferation, or apoptosis depending on conditions. It is widely used as a phenotypic marker for mouse T cells.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.5 µg/test (in 100 µL final volume). Optimal antibody concentration should be titrated for each assay.

Target Information

The CD3 complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, all encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 plays a role in TCR-induced growth arrest, survival, and proliferation. It is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
