Product Specific Information

The eBioG4.18 monoclonal antibody recognizes the rat CD3 protein, a critical component of the T-cell receptor (TCR) complex and a marker of T-cell lineage. CD3 participates in signal transduction from the TCR to the nucleus, leading to transcription of IL-2 and clonal expansion.

• In vitro: Immobilized G4.18 antibody activates T cells; soluble G4.18 inhibits allogeneic mixed-lymphocyte proliferation and cytotoxicity.
• In vivo: Demonstrated to induce long-term specific tolerance to organ allografts.

Applications Reported: Flow cytometry, immunoblotting (WB), immunohistochemistry, and functional assays.
Applications Tested: Flow cytometric analysis of rat splenocytes (≤0.5 µg/test, in 100 µL final volume). Cell number should be optimized (10⁵–10⁸ cells/test). Antibody titration is recommended for best results.

Target Information

CD3 is a subunit complex essential for transducing antigen-recognition signals into T cells and regulating TCR complex expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on human chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs that interact with signaling molecules.

• Expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells.
• Not expressed on B or NK cells.
• Decreased T lymphocyte percentages may be observed in autoimmune diseases.
• CD3 gene defects are associated with CD3 immunodeficiency.
