Description:
The eBio500A2 monoclonal antibody reacts with the 25 kDa epsilon subunit of the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells and NKT cells. Binding of eBio500A2 to CD3 initiates intracellular signaling leading to cellular activation and proliferation. The 500A2 antibody partially cross-blocks the 17A2 and 145-2C11 antibodies, indicating recognition of distinct but overlapping epitopes.

Applications Tested:
This antibody has been tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended use is ≤0.5 µg per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test. Optimal dilution should be determined experimentally.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 gamma, delta, epsilon, and zeta subunits are structurally related immunoglobulin superfamily members encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via a double tyrosine-based motif. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may occur in some autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
