Target Information
NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and p53 regulation.
NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones. It binds nucleic acids, prevents protein aggregation, protects enzymes during thermal denaturation, and assists renaturation of denatured proteins.
NPM1 is associated with centrosome duplication and acts as a substrate of CDK2/cyclin E. It participates in tumor-associated fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, functioning as an interface for oligomerization.
Dysfunction of NPM1 is linked to diseases including acute myeloid leukemia and lymphomatoid papulosis.
