Immunogen Clone M415 was generated from phospho-CREB (Ser-133) synthetic peptide (coupled to carrier protein) corresponding to amino acids surrounding Ser-133 in human CREB. This sequence is well conserved in rat, mouse, chicken, and zebrafish CREB. The site is also conserved in CREM (Ser-136) and ATF1 (Ser-63).

Product Specific Information

This antibody detects a 43 kDa protein corresponding to the molecular mass of CREB on SDS-PAGE immunoblots of human A431, Jurkat, and rat PC12 cells treated with Calyculin A, as well as A431 treated with EGF and PC12 treated with NGF or Forskolin. This reactivity is not observed after lambda phosphatase treatment.

Target Information

CREB (Cyclic AMP response element binding protein) is a 43 kDa basic/leucine zipper transcription factor that binds the cyclic AMP response element (CRE) and activates transcription in response to extracellular signals such as neurotransmitters, hormones, membrane depolarization, and growth factors. Activation of CREB depends on phosphorylation at serine 133, mediated by kinases such as p44/42 MAPK, p90RSK, p38 MAPK, and MSK1. While CREB can bind DNA regardless of phosphorylation, only the phosphorylated form acts as an active transcription factor. CREB interacts with CREB binding protein (CBP) at serine 133 and is involved in processes including circadian rhythm regulation, adipocyte differentiation, and learning and memory. The human CREB gene is located on chromosome 2q. Dysregulation of CREB function is associated with diseases such as Alzheimer’s disease, histiocytoma, and soft tissue melanoma.
