In Western blot applications, this antibody detects bands at approximately 35 and 45 kDa in non-apoptotic cells. Jurkat cell lysate is recommended as a positive control.

Target Information

ICAD (Inhibitor of Caspase-Activated DNase) regulates nuclear apoptosis by inhibiting CAD (Caspase-Activated DNase).
It has two isoforms:

• ICAD-L/DFF45 (45 kDa) – functional isoform
• ICAD-S/DFF35 (35 kDa) – non-functional isoform

ICAD is cleaved by caspase-3 at residues 117 and 224, releasing fragments that activate CAD to fragment nuclear DNA during apoptosis.
Beyond its inhibitory role, ICAD serves as a folding chaperone for CAD.
ICAD is considered a potential therapeutic target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified.
