Takara Lenti-X Maxi Purification Kit

The Lenti-X Maxi Purification Kit produces outstanding yields of highly purified lentivirus from crude supernatants. The gravity column-based protocol is fast, simple, and effective, offering superior performance compared to filter-based purification systems that can damage fragile lentiviral particles and reduce yields. The gravity-flow column gently retains virus particles from the supernatant, while unbound material flows through during the wash steps. Eluted virus is recovered fully intact and functional—purifying virus is now as simple as purifying plasmid DNA.

Why Purify Your Lentivirus?

Lentivirus purification removes cellular contaminants that can negatively affect transduction experiments. Crude supernatants contain residual plasmid DNA, infection and transduction inhibitors, cellular and serum proteins, and immunogenic viral proteins, nucleic acids, and virus fragments.
Before using lentivirus with sensitive target cells or for in vivo applications, purification is essential to eliminate these contaminants.

Simple Procedure

• Prepacked, ready-to-use purification columns
• Add 10X binding buffer to your supernatant sample (9–45 ml)
• Apply to the column and allow gravity flow
• Impurities are removed during two wash steps
• Purified lentivirus is recovered in 3 ml of elution buffer

Prevent Pseudotransduction

Purified lentivirus stocks prevent pseudotransduction, where recombinant proteins in crude supernatant are passively transferred to target cells. This ensures the observed expression originates from de novo transduction rather than contamination.

Higher Yield and Purity

A gentle purification procedure maintains viral infectivity and achieves excellent yields of 60–80%.
In contrast, anion exchange-based membrane systems yield less than 20%.
Lenti-X columns produce virus preparations with minimal extraneous protein contamination, achieving significantly higher purity than filter-based methods.
