- In Western blot, multiple bands may appear at 100–120 kDa, representing post-translational modification of HIF-1α.
- For WB, testing on nuclear extracts is recommended.
Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 consists of alpha and beta subunits belonging to the PAS family. It regulates transcriptional responses to hypoxia, activating genes such as erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha plays roles in apoptosis, cell proliferation, and tumor angiogenesis under hypoxic conditions. It interacts with p53 and MDM2 for degradation regulation and contributes to ROS maintenance via NOX4 induction. Located on chromosome 14q, its C-terminal binds to p300/CBP complexes to induce hypoxia-responsive genes.
Hypoxia-related mechanisms involving HIF1A are implicated in ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
