- In Western blot, multiple bands may appear at 100–120 kDa, representing post-translational modifications of HIF-1α.
- For WB, testing on nuclear extracts is recommended.

Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family of transcription factors.
HIF1 acts as a transcriptional regulator of the adaptive response to hypoxia, activating over 40 genes such as erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 and p300/CBP complexes to modulate hypoxia-responsive gene expression.
Located on chromosome 14q in humans, HIF1A plays a critical role in diseases such as ischemia, cancer, pulmonary hypertension, and chronic obstructive pulmonary disease.
