Target Information
Thioredoxins (Trx) are small, multifunctional proteins with oxidoreductase activity and are ubiquitous in almost all living cells. Trx contains a redox-active disulfide/dithiol group within the conserved Cys-Gly-Pro-Cys active site. The two cysteine residues can form intramolecular disulfide bonds, which are reduced by Trx reductase using NADPH as an electron donor.
Reduced Trx serves as a hydrogen donor for ribonucleotide reductase, essential for DNA synthesis, and acts as a general protein disulfide reductase involved in growth and redox regulation. Trx also reduces protein disulfide-isomerase (PDI) and transcription factors such as p53, NF-kB, and AP-1. Furthermore, Trx is capable of removing H₂O₂ when coupled with methionine sulfoxide reductase or peroxiredoxins.
