Product Specific Information
This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin of the Ig superfamily that binds sialic acid–containing glycoconjugates. Siglec F is mainly expressed on eosinophils and alveolar macrophages, with lower expression on immature myelomonocytic cells. It is the functional ortholog of human Siglec 8 but is not expressed on mast cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on thioglycolate-elicited peritoneal macrophages; ≤0.125 µg/test recommended. Adjust cell number empirically (10⁵–10⁸ cells/test).
Light Sensitivity: Protect from light due to tandem dye photo-oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using:
Excitation: 488–561 nm
Emission: 775 nm
Compatible Lasers: Blue, Green, Yellow-Green
Target Information
SIGLEC5 is a putative adhesion molecule mediating sialic acid–dependent binding to cells. It binds both α2,3- and α2,6-linked sialic acids. Expression is high in peripheral blood leukocytes, spleen, and bone marrow, and lower in lymph node, lung, appendix, placenta, pancreas, and thymus. It is expressed by monocytes and neutrophils but absent in leukemic cell lines representing early myelomonocytic differentiation stages.
