Immunogen Clone M132 was generated from a phospho-GSK-3β (Tyr-216) synthetic peptide (coupled to KLH) corresponding to amino acid residues around tyrosine 216 of human GSK-3β. This peptide sequence is also found in GSK-3α (Tyr-279) and is highly conserved in GSK-3 genes in rat and mouse.

The antibody detects 46/50 kDa proteins corresponding to the apparent molecular mass of GSK-3β and GSK-3α on SDS-PAGE immunoblots of pervanadate-treated rabbit fibroblasts, as well as treated human SKN-SH and A431 cells.

Target Information

Glycogen synthase kinase-3 (GSK3) is a protein kinase originally identified as a regulator of glycogen synthase, a key enzyme in glycogen metabolism. It regulates diverse cellular functions such as protein synthesis, cell proliferation, differentiation, microtubule assembly/disassembly, and apoptosis.
GSK3’s substrate specificity requires a pre-existing phosphoserine or phosphotyrosine four residues C-terminal to the phosphorylation site.
Two isoforms, GSK3α and GSK3β, are regulated by phosphorylation. Inactivation occurs via Ser9 (Ser21 in GSK3α) phosphorylation by PKB, while activation depends on Tyr216 (Tyr279 in GSK3α) phosphorylation.
Activated GSK3β phosphorylates multiple cellular proteins including p53, c-Myc, c-Jun, HSF-1, and cyclin D1, and is implicated in Alzheimer’s disease progression through tau phosphorylation.
