Product Specific Information

This antibody detects a 38 kDa protein on SDS-PAGE immunoblots of Jurkat cells treated with calpain inhibitor (ALLN) followed by TNFα. The antibody does not detect this band in untreated cells.

Target Information

IkB-alpha is a 40 kDa protein that inhibits NF-κB activity through protein-protein interaction with NF-κB dimers in the cytosol, masking their nuclear localization sequence and preventing nuclear translocation. Various stimuli (TNF-α, IL-1β, chemokines, PMA, growth factors, LPS, UV, viral infection, and stress) induce NF-κB activation by promoting IkBα degradation.

Phosphorylation of IkBα on Ser32 and Ser36 by IKKα triggers its degradation. IKKα (CHUK) interacts with IkBα and is essential for NF-κB activation in response to proinflammatory cytokines. The IKK complex (IKKα, IKKβ, IKKγ) is a key regulator of this phosphorylation process.

In humans, the IkBα gene is located on the q arm of chromosome 14.
