Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. The STAT4 protein shares 52% similarity with STAT1 and 47% with STAT3. Like other STAT family members, it undergoes tyrosine phosphorylation by Jak1 or Jak2, forming homo- and heterodimers. Tyrosine-phosphorylated STAT4 binds to the IFN-gamma activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity.
STAT4 expression is mainly found in the thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which is critical for IFN-gamma induction and Th1 response development. STAT4-deficient mice show impaired Th1 and enhanced Th2 differentiation. Additionally, IFN-α/β activation of STAT4 is required for IFN-γ production during viral infection.
