Target Information

This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid residue separating the first two of four conserved cysteine residues.

Binding of chemokines to CXCR3 induces cellular responses such as leukocyte trafficking, integrin activation, cytoskeletal rearrangement, and chemotactic migration. Pertussis toxin inhibition indicates coupling with Gi-type G proteins. The receptor undergoes phosphorylation-dependent internalization upon activation, leading to rapid but transient responses, followed by receptor recycling.

CXCR3 is highly expressed in effector/memory T cells and inflamed tissues, playing a key role in immune cell recruitment during inflammation. Consequently, it is a potential target for small-molecule antagonists in inflammatory disease treatment. Multiple transcript variants encoding different isoforms have been identified.
