Target Information
Neurofilaments (NFs) are intermediate filaments expressed almost exclusively in neuronal cells, prominently in large axons. NFs are composed of three polypeptide chains—NF-M, NF-H, and NF-L—which share sequence and structural similarity in the coiled-coil core domain but differ in terminal regions. NF-M and NF-H have flexible C-terminal extensions that link neurofilaments to other cytoplasmic elements. NF-M tail-mediated interactions are crucial for axonal size and cytoskeletal architecture. Phosphorylation and O-GlcNAcylation of NF-M are reciprocally regulated, affecting filament formation and function. Antibodies to neurofilament subunits serve as robust neuronal markers due to their abundance and stability in the nervous system.
