Product Specific Information

Description:
The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1 in human, and OX-45 in rat). CD48 is a 45 kDa GPI-linked glycoprotein, a member of the SLAM and immunoglobulin superfamily, expressed on most hematopoietic cells. It plays a role in adhesion and T cell activation.

CD48 expression varies among hematopoietic progenitors:

• HSC: CD150(+)CD244(-)CD48(-)
• MPP: CD244(+)CD150(-)CD48(-)
• Restricted progenitors: CD48(+)CD244(+)CD150(-)

HM48-1 modulates CD48 functions by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-activated spleen cell proliferation, providing costimulatory signals for T cells, and prolonging cardiac allograft survival in vivo.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤ 0.2 µg/test (100 µL final volume; 10⁵–10⁸ cells/test). Optimize titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD48 (Blast-1) is part of the CD2 subset of the Ig superfamily, including CD2, CD58, CD84, CD150, CD229, CD244, and others. These molecules mediate homotypic or heterotypic adhesion.

• Expression: Broadly on hematopoietic cells; strong on lymphocytes and monocytes, weak on granulocytes, absent on platelets, fibroblasts, epithelium, and endothelium.
• Function: Serves as a ligand for CD2 and 2B4; regulates NK and NK-T cell proliferation; involved in eosinophil activation and immune responses.
• Clinical relevance: Marker for detecting GPI anchoring defects in PNH; potential target for allergic disease therapies (e.g., rhinitis, asthma).
