Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen, also known as BCM1, Blast-1 (human), and OX-45 (rat).
CD48 is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells and is part of the SLAM family. It is useful for predicting the primitiveness of hematopoietic progenitors based on expression of SLAM family members.

• MPP: CD244(+) CD150(-) CD48(-)
• Restricted progenitors: CD48(+) CD244(+) CD150(-)

CD48 is involved in adhesion and T cell activation. In mice, its primary counter-receptors are CD2 and CD244. HM48-1 modulates CD48 functions by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting spleen cell proliferation, providing costimulation for T cells, and prolonging cardiac allograft survival.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample or per 100 µL cell sample.

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow Laser).

Note: Internal testing shows NovaFluor Yellow 610 non-specifically stains B cells in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily, including CD2, CD58, CD84, CD150, CD229, CD244, etc.
It mediates adhesion via homotypic or heterotypic interactions. CD48 is a GPI-anchored protein expressed on hematopoietic cells and serves as a ligand for 2B4 and CD2.

• Regulates NK/NK-T cell proliferation via 2B4-CD48 interaction
• Involved in eosinophil activation and infection responses
• Expressed strongly on lymphocytes and monocytes, weakly on granulocytes
• Absent on platelets, fibroblasts, epithelium, and endothelium
• Marker for detecting GPI anchoring defects (e.g., PNH)
• Potential pharmacological target for allergic airway diseases (rhinitis, asthma)
