Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1, and OX-45).
CD48 is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells and belongs to the SLAM and Ig superfamilies. It plays a key role in adhesion and T-cell activation.

• Expression in hematopoietic progenitors:
  • HSC: CD150(+)CD244(-)CD48(-)
  • MPP: CD244(+)CD150(-)CD48(-)
  • Restricted progenitors: CD48(+)CD244(+)CD150(-)

HM48-1 modulates CD48 functions by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-activated spleen cell proliferation, providing costimulation for T cells, and prolonging cardiac allograft survival in vivo.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

• Recommended to titrate for optimal assay performance

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining antibody with cells
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CD48 (Blast-1) is a GPI-anchored protein of the CD2 subset of the Ig superfamily (including CD2, CD58, CD84, CD150, CD229, CD244, etc.).
It mediates adhesion through homotypic or heterotypic interactions and serves as:

• High-affinity ligand for 2B4
• Low-affinity ligand for CD2

Expression profile:

• Strong: lymphocytes, monocytes
• Weak: granulocytes
• Absent: platelets, fibroblasts, epithelium, endothelium

CD48 is involved in NK and NK-T cell proliferation, eosinophil activation, and is a marker for GPI anchoring defects (e.g., PNH).
It may also be implicated in allergic airway diseases, offering potential therapeutic targets.
